Background: Alzheimer's disease (AD) and diabetes are epidemiologically associated, but their shared genetic mechanisms remain unclear.
Methods: We investigated the shared genetic architecture between AD and diabetes-related metabolic traits using two-sample Mendelian randomisation, linkage disequilibrium score regression (LDSC), local genetic correlation (LAVA), gene overlap analysis, and pathway enrichment.
Results: No evidence of a causal relationship was observed. However, significant global and local genetic correlations were identified between AD and several metabolic traits, including glucose, insulin, and HbA1c. Shared genome-wide significant genes were enriched in lipid metabolism and immune-related pathways.
Conclusion: AD and diabetes-related metabolic traits share common genetic architecture and biological pathways, providing insights into disease mechanisms and potential therapeutic targets.
