A global, diverse cohort study of over 1,200 early-onset Alzheimer's cases revealed that known genetic variants (APP, PSEN1/2) account for only 1.44% of cases, indicating a high prevalence of unknown genetic drivers. Results highlight that the majority of early-onset Alzheimer’s disease is idiopathic, requiring broader genome-wide discovery to identify novel, ancestry-specific risk factors.
