Correlation Between Csvd Neuroimaging Biomarkers And Plasma Plgf Among Ad And Non-ad Participants In The Memory Clinic Cohort From Thailand

Part of Topic

Background:

Cerebral small vessel disease (CSVD) is one of the main contributing factors to vascular cognitive impairment and dementia (VCID) in the aging population. CSVD can contribute to cognitive decline by itself or worsen cognitive decline as a co-pathology in Alzheimer’s disease (AD). Plasma placental growth factor (PlGF), a member of the vascular endothelial growth factor family, is a promising novel biomarker of subcortical white matter burden in both AD and non-AD populations. This study investigates the correlation between CSVD neuroimaging biomarkers and plasma PlGF in a memory clinic setting.

Methods:

The clinical data and plasma biomarkers were collected from memory clinic cohorts. AD and non-AD groups were defined by an abnormal cerebrospinal fluid Amyloid beta (Aβ) 42/p-tau181 ratio, or a positive Aβ Positron Emission Tomography. Magnetic resonance imaging (MRI) was independently rated by the two trained investigators using STRIVE-2 criteria (Duering M, 2023). The validated composite total CSVD score (Staals J, 2014), ranging from 0-4 was calculated. The non-parametric statistical analysis was applied to analyze.

Results:

A total of 114 participants were recruited. Clinical information, laboratory test results, and neuroimaging biomarkers were compared between AD and non-AD groups, as shown in Table 1. Plasma PlGF did not show a statistically significant difference between AD and non-AD groups. The Spearman’s correlation and the Wilcoxon rank sum test between plasma PlGF and each CSVD neuroimaging biomarker are summarized in Table 2. Plasma PlGF did not show a statistical correlation with CSVD scores or any of the CSVD neuroimaging biomarkers.

Conclusion:

Plasma PlGF showed similar values between AD and non-AD groups. Neither CSVD biomarkers nor total CSVD score correlated with plasma PlGF. These findings showed a limitation of measuring plasma PlGF as a solitary biomarker of CSVD burden in both AD and non-AD participants in a memory clinic setting. Further evaluation of the relationship between PlGF and quantitative volumetric white matter hypertensities should be explored.
