Use Of Chemically Peptide-ligated Protein Standards For High Sensitivity Detection Of Phosphorylated Tau In Alzheimer's Disease Samples

Part of Topic

Accurate quantification of phosphorylated Tau (pTau) is critical for advancing Alzheimer’s disease (AD) biomarker research, yet generating suitable standards and highly specific antibodies for post-translational modifications remains challenging. Here, we describe an approach combining chemically peptide-ligated protein (CLP) standards with SimpleStep® ELISA technology to enable sensitive and specific detection of pTau epitopes.

CLPs were generated by ligating synthetic phosphopeptides to recombinant proteins, providing well-defined standards for assay development and antibody screening. A structured, three-step workflow was applied to identify phospho- and sequence-specific rabbit monoclonal antibodies targeting Tau phosphorylation sites (T181, S199, T217, T231, and S396). This strategy enabled efficient screening for specificity, cross-reactivity, and performance in both recombinant systems and biological samples, resulting in the selection of optimal antibody pairs.

Validated SimpleStep® colorimetric ELISA kits demonstrated robust performance across multiple sample types, meeting established criteria for precision, recovery, and linearity. Application to human brain tissue extracts revealed measurable differences in phosphorylated Tau levels between normal and AD samples. Furthermore, integration of the SimpleStep Ignite™ chemiluminescent platform significantly enhanced assay sensitivity (up to ~16-fold), enabling detection of low-abundance pTau in biofluids including serum, plasma, and CSF.

Overall, this work demonstrates that CLP-based standards combined with streamlined antibody screening support the development of high-performance ELISA assays for pTau quantification. These tools provide improved sensitivity and specificity for studying Tau pathology and have potential utility in both translational research and clinical biomarker applications.
