Background: The CenTauR project provides universal measurement scales and regions-of-interest (ROIs) for standardizing tau PET quantification units across tracers and pipelines. In 2024, we released two fully automated software pipelines on the Global Alzheimer’s Association Interactive Network (GAAIN) CenTauR website (https://www.gaain.org/centaur-project) to compute numeric CenTauRZ and CenTauR SUVR values for input pairs of T1-w MRI and late-uptake summed tau PET NIfTI files from 6 different tracers: FTP (AV-1451), MK-6240, RO948, GTP1, PI2620, or PM-PBB3, with 5 different CenTauR regions of interest (ROIs): universal (global), mesial temporal, meta-temporal, temporoparietal, and frontal. One was based on SPM8, mirroring the Centiloid Standard approach. The other is a more modern approach that quantifies images in MRI space incorporating SPM12 tissue-class segmentations and the Mayo Clinic Adult Lifespan Template. Both automatically apply included calibration equations.
In 2025, the CenTauR group have developed new calibration (slope and intercept) equations for “CenTauR” units, determined using their Joint Propagation Model (JPM) (Leuzy et al., 2024). Unlike CenTauRZ, CenTauR units are designed to allow quantitative comparison of continuous values across tracers at all levels of pathology, analogous to the typical usage of Centiloid units for amyloid PET. The SPM12-based pipeline was also adopted as the CenTauR standard software pipeline, which is used to calibrate all other pipelines.
Methods: We have updated the GAAIN.org CenTauR software with the new “CenTauR” calibration equations to now additionally output values in “CenTauR” units. The older SPM8-based pipeline has now been deprecated, along with CenTauRZ units.
Results: The updated pipeline will be released at https://www.gaain.org/centaur-project when the CenTauR publication is accepted.
Discussion: The CenTauR pipeline allows researchers all over the world to easily quantify tau PET scans in CenTauR standardized units, and to calibrate other pipelines to this standard, enabling greater harmonization across tracers and across research groups.
