INTRODUCTION

▪ Alzheimer’sdisease(AD)islinkedtotheearlybuildupofamyloid-beta(Aβ)inthebrainbeforecognitiveproblemsappear.
▪ Clinicalevidenceshowsthatamyloid-positiveindividualsexhibitasignificantlyfasterrateofdeclineinepisodicmemorythanamyloid-negativeindividuals.
▪ Deficitsinassociativememoryanddelayedrecallaredeeplylinkedtoamyloiddeposition,distinguishingthemfromthemoregradualchangesseeninnormalaging.

METHODS

- Qualitativemethodologywasusedtoobtaininformationfromrelevantliteratureonthemeaningandrolesofamyloidinmemoryloss.
▪ Datawereanalysedusingsystematicreviewtoexplaintheprevalenceandoutcomesofamyloid-positiveandamyloid-negativeinrelationtomemoryloss

RESULTS

- Alzheimer’sdiseasestartswhenabnormalproteinsbuildupinthebrain,formingamyloidplaquesandneurofibrillarytangles.
▪ Thoughamyloid-beta(Aβ)normallyhelpswithbrainfunction,toomuchbuildupofitshowsanearlysignofAlzheimer’sdisease(AD).
▪ Thechangefromamyloid-betanegative(Aβ−)toamyloid-betapositive(Aβ+)showstheprogressionoftheAD,butthefactorsthatpredictthischangearestillnotclinicallyclear..
▪ Aβ+islinkedtoafasterrateofbraintissueloss(atrophy)whencomparedtoAβ-.
▪ ClinicalStudiesshowthatamongindividualswithoutdementia,amyloidpositivityismoreprevalentwithanincreasedageandismorefrequentinwomenthaninmen

CONCLUSIONS

▪ IndividualsofAβ+andAβ-showmemorydecline,butthosewithamyloid-positiveexperienceafasterandmoreseveredecline,especiallyinassociativememoryanddelayedrecall.
▪ Addressingmemorylossrequiresamultimodalapproachthatwillincorporatesensitivecognitiveassays,structuralMRI,andcoreADbiomarkersforaccuratedifferentiationinrefiningprognosticaccuracy,enabletimelyintervention,andensuretheappropriateallocationofindividualsintoclinicaltrialsandpreventativecarepathways
