LNP technology enabled efficient and reproducible isolation of EVs from human plasma, yielding material suitable for downstream multi-omic analyses. Using the NULISA platform, multiple CNS- and AD-relevant proteins were robustly detected in LNP-isolated EVs, including amyloid-β 40 (Aβ40), amyloid-β 42 (Aβ42), phosphorylated tau species (pTau-181, pTau-217, pTau-231), phosphorylated TDP-43 (pTDP43-409), and glial fibrillary acidic protein (GFAP). Consistent with enrichment of CNS-derived EV cargo, neuronal-associated transcripts such as DLG1, GAD1, MAP2, NRGN, NRXN, and NEUROD1 were also detected by RT-qPCR.
